Biological Background: NF-kB p65/RelA Function and Localization
- Transcription factor p65, also known as RELA, nuclear factor NF-kappa-B p65 subunit, encoded by the RELA gene (NFKB3).
- NF-kappa-B is a pleiotropic transcription factor present in almost all cell types, involved in inflammation, immunity, differentiation, cell growth, tumorigenesis and apoptosis.
- Forms homo- or heterodimers with Rel domain proteins; the heterodimer with NFKB1/p50 is the most abundant, acting as a transcriptional activator.
- Subcellular location is both nucleus and cytoplasm; inactive complexes are retained in the cytoplasm via interaction with I-kappa-B inhibitors, and upon activation, translocate to the nucleus.
- Subject to multiple post-translational modifications including acetylation, phosphorylation, methylation, and ubiquitination, which regulate its activity.

Experimental Guidance and Technical Tips
- Consider optimizing antibody dilution for each application and sample type. For Western blot, a band at approximately 60 kDa is expected.

- In immunocytochemistry, fixation and permeabilization conditions may influence staining; inclusion of appropriate controls is recommended.
- The antibody may be suitable for ELISA, but pairing with a detection antibody should be empirically determined.
