Biological Background: NF-kB p65/RelA Function and Localization

  • Transcription factor p65, also known as RELA, nuclear factor NF-kappa-B p65 subunit, encoded by the RELA gene (NFKB3).
  • NF-kappa-B is a pleiotropic transcription factor present in almost all cell types, involved in inflammation, immunity, differentiation, cell growth, tumorigenesis and apoptosis.
  • Forms homo- or heterodimers with Rel domain proteins; the heterodimer with NFKB1/p50 is the most abundant, acting as a transcriptional activator.
  • Subcellular location is both nucleus and cytoplasm; inactive complexes are retained in the cytoplasm via interaction with I-kappa-B inhibitors, and upon activation, translocate to the nucleus.
  • Subject to multiple post-translational modifications including acetylation, phosphorylation, methylation, and ubiquitination, which regulate its activity.
