Biological Background: NF-kB p65/RelA Function and Localization
- NF-kB p65/RelA is a subunit of the NF-kB transcription factor complex, which exists as homo- or heterodimers; the heterodimer with NFKB1 is the most abundant form and acts as a transcriptional activator.
- It plays a central role in inflammation, immunity, differentiation, cell growth, tumorigenesis, and apoptosis through regulation of target gene expression.
- NF-kB activation involves phosphorylation and degradation of I-kB inhibitors, leading to the release and nuclear translocation of the active p65/RelA complex.
- Essential for cytokine gene expression in T-cells. Related references: PMID:15790681
- Key transcription factor regulating the interferon response during SARS-CoV-2 infection. Related references: PMID:33440148
- Associates with chromatin at NF-kB promoter regions via interaction with DDX1, modulating promoter accessibility.
- Subject to multiple post-translational modifications including acetylation, phosphorylation, methylation, and ubiquitination, which fine-tune its activity.
- Predominantly cytoplasmic in resting cells, translocates to the nucleus upon activation; also found in the nucleus under basal conditions.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a synthetic peptide from the C-terminal region (aa 450-551) of human NF-kB p65. For applications like WB, expect a band around 58-60 kDa; consider using appropriate loading controls and validating the signal in KO cell lines.
- This antibody is suitable for multiple applications; for IF/ICC, both nuclear and cytoplasmic signals may be observed depending on the activation state of NF-kB. Include untreated and stimulated samples for comparison.
- For ChIP assays, optimize crosslinking and sonication conditions for your cell type. The antibody's performance should be validated with positive and negative target gene regions.
- Cross-reactivity with mouse, rat, and monkey is reported; always verify specificity in your species and tissue of interest before scaling up experiments.
- When using this KO-validated antibody, consider including a negative control (e.g., siRNA knockdown or CRISPR KO lysate) to confirm band specificity in WB.
