- Nexilin (NEXN), also known as Nelin or CMH20, is an F-actin-binding protein that plays a critical role in the maintenance of Z-line and sarcomere integrity in striated muscle.
- It regulates cell migration by directly associating with the actin cytoskeleton, influencing cytoskeletal dynamics and adhesion.
- The protein contains immunoglobulin-like domains and undergoes alternative splicing, producing multiple isoforms with tissue-specific distributions.
- Subcellularly, NEXN localizes to the cytoplasm, cytoskeleton, cell–cell adherens junctions, and the sarcomeric Z-line of myofibrils.
- Post-translational modifications include phosphorylation (phosphoprotein), which may modulate its actin-binding activity or localization.

- For Western blot, a band near 81 kDa is expected in heart and skeletal muscle lysates; validate the antibody in these tissues along with a negative control (e.g., NEXN knockout or siRNA-treated cells).
- In immunofluorescence/ICC, fixation and permeabilization conditions should be optimized to preserve the delicate sarcomeric Z-line and junctional structures. Co-staining with actin or Z-line markers may confirm localization.
