Biological Background: Neutrophil Elastase (ELANE) Function and Localization
- ELANE is primarily expressed in bone marrow cells and neutrophils (PubMed:10947984).
Related references: PMID:10947984 - It modifies the functions of natural killer cells, monocytes, and granulocytes; inhibits C5a-dependent neutrophil enzyme release and chemotaxis (PubMed:15140022).
Related references: PMID:15140022 - Exhibits microbicidal activity by mediating the cleavage and degradation of bacterial proteins; kills E. coli but not S. aureus in vitro; digests outer membrane protein A (OmpA) in E. coli and K. pneumoniae (PubMed:10947984, PubMed:25161283).
Related references: PMID:10947984, PMID:25161283 - Critical for the formation of neutrophil extracellular traps (NETs): translocates to the nucleus downstream of MPO and degrades histone H4, promoting chromatin decondensation (PubMed:20974816, PubMed:25066128, PubMed:28574339).
Related references: PMID:20974816, PMID:25066128, PMID:28574339 - Promotes blood coagulation and platelet aggregation by inactivating tissue factor pathway inhibitor (TFPI) and activating the fibrinogen receptor integrin alpha-IIb/beta-3 (PubMed:20676107, PubMed:25211214, PubMed:9111081).
Related references: PMID:20676107, PMID:25211214, PMID:9111081 - Inhibits pyroptosis by promoting the cleavage of GSDMB (PubMed:36899106).
Related references: PMID:36899106 - Subcellular localization includes cytoplasmic vesicles, phagosomes, lysosomes, and cytolytic granules; actively translocates to the nucleus during NETosis.
- Synthesized as a glycosylated preproenzyme; activated by proteolytic removal of the propeptide; stabilized by disulfide bonds.
Experimental Guidance and Technical Tips
- For Western blotting, the mature enzyme has a predicted molecular weight of ~29 kDa; however, the proform and glycosylation may lead to higher bands. Validate the observed band with appropriate controls.
- For immunohistochemistry (IHC) and immunofluorescence (IF), neutrophil-rich tissues such as bone marrow or inflamed tissue sections are recommended as positive controls. Note that ELANE can be secreted and may show diffuse staining.
- When using for ELISA, consider this polyclonal antibody as a capture or detection reagent. Pairing with a monoclonal antibody against a non-overlapping epitope may improve assay specificity.
