Biological Background: Neutrophil Elastase (ELANE) Function and Localization

• Neutrophil elastase (ELANE), also known as bone marrow serine protease, elastase-2, human leukocyte elastase (HLE), medullasin, and PMN elastase, is a serine protease encoded by the ELANE gene.

• The immunogen is a synthetic peptide from the catalytic domain (aa 167-267), suggesting the antibody likely recognizes the mature active form of ELANE. Validate specificity using recombinant protein or cell lysates expressing active ELANE.
• For Western blot (WB), the predicted molecular weight is ~29 kDa; note that the unprocessed zymogen may appear at a higher molecular weight. Confirm band identity with appropriate controls.
• In immunoprecipitation (IP) experiments, consider the antibody's potential preference for the active enzyme form, which is relevant for studying protease interactions and complexes.
• For ELISA applications, the antibody may be used as a capture or detection reagent. Consider pairing with another ELANE-specific antibody for sandwich ELISA development.
