Biological Background: Neuropilin-1 Function and Localization
- Neuropilin-1 (NRP1) is a transmembrane receptor also known as Vascular endothelial cell growth factor 165 receptor. It mediates semaphorin-dependent chemorepulsion and vascular endothelial growth factor (VEGF)-induced angiogenesis, playing crucial roles in cardiovascular development and neuronal circuit formation. Related references: PMID:10688880 PMID:9288753 PMID:9529250
- NRP1 recognizes a C-end rule (CendR) motif (R/KXXR/K) on ligands such as VEGF165 and semaphorin 3A, facilitating cellular internalization and vascular permeability. Co-expression with KDR/VEGFR2 enhances VEGF165 binding and chemotaxis. Related references: PMID:19805273
- The protein regulates mitochondrial iron transport via interaction with ABCB8/MITOSUR, linking it to cellular metabolism. Related references: PMID:30623799
- NRP1 acts as a host factor for SARS-CoV-2 infection by binding the CendR motif RRAR on the spike protein, enhancing viral entry.
- Subcellularly, NRP1 is found at the cell membrane, cytoplasm, and mitochondrion membrane; it is also secreted. Keywords include angiogenesis, neurogenesis, host-virus interaction, and alternative splicing.
- Tissue expression is prominent in the developing nervous system, heart, and placenta; in adults, it is high in heart and placenta, moderate in lung, liver, kidney, and low in brain. Isoforms 1 and 2 show non-overlapping patterns. Related references: PMID:33082293 PMID:36213313 PMID:10688880 PMID:9529250
- Soluble NRP1 isoforms may bind VEGF165 and inhibit its activity, potentially inducing apoptosis by sequestering VEGF165. Related references: PMID:10748121 PMID:26503042
- Post-translational modifications include glycosylation, heparan sulfate attachment, and phosphorylation, consistent with its roles in signaling and regulation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal cytoplasmic domain (aa 824-923), suggesting the antibody may detect both membrane-bound and soluble NRP1 isoforms in Western blotting.
- For WB, expected band size is ~103 kDa (full-length); consider using reducing conditions and protease inhibitors.
- For IF/ICC, validate fixation/permeabilization protocols to preserve antigen accessibility.
- Cross-reactivity with mouse and rat is reported, making it suitable for translational studies; confirm reactivity in relevant tissue lysates.
- ELISA application may be used for quantification of NRP1 in serum or cell supernatants; titrate antibody concentration for optimal signal-to-noise.
