Biological Background: Neuropilin-1 Function and Localization
- Neuropilin-1 (NRP1) is a multi-functional cell-surface receptor also known as Vascular endothelial cell growth factor 165 receptor, CD304, and BDCA4. Alternative splicing yields isoforms with non-overlapping expression patterns, contributing to its diverse tissue-specific functions.
- It binds class 3 semaphorins (e.g., SEMA3A) and VEGF family members (VEGF165, PLGF-2, VEGFB), mediating chemorepulsion and angiogenesis, respectively. Related references: PMID:10688880, PMID:9288753, PMID:9529250
- NRP1 recognizes a C-end rule (CendR) motif (R/KXXR/K) on ligands, triggering cellular internalization and vascular leakage. Related references: PMID:19805273
- Coexpression with VEGFR2/KDR enhances VEGF165 binding and downstream signaling, promoting endothelial cell chemotaxis and angiogenesis.
- In the developing nervous system, NRP1 guides motor neuron axon projection and cell body migration, including facial motor neuron migration from rhombomere 4 to 6.
- NRP1 localizes to the cell membrane, cytoplasm, and is also found secreted and associated with the mitochondrial membrane, reflecting its multifunctional roles.
- In adult tissues, NRP1 is highly expressed in heart and placenta, moderate in lung, liver, skeletal muscle, kidney and pancreas, and low in brain. Related references: PMID:10688880, PMID:9529250
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 824–923 within the C-terminal domain of human NRP1. This region is conserved in mouse and rat, consistent with the reported cross-reactivity. Verify sequence alignment in your species of interest prior to experiments.
- For Western blot (WB) analysis, a band near 103 kDa is expected for full-length NRP1. Consider using membrane protein extraction protocols and reducing conditions, as NRP1 forms disulfide-linked dimers and may exhibit higher-order structures. Validate specificity with appropriate controls (e.g., peptide competition or knockdown).
- For ELISA, this antibody may be suitable as a capture or detection reagent; titrate conditions using recombinant NRP1 or known positive samples and include negative controls to establish assay sensitivity and dynamic range.
- Additional bands may appear due to isoform heterogeneity, glycosylation, or proteoglycan modifications; confirm band identity using mass spectrometry or isoform-specific reagents if needed.
- Store at -20°C or lower upon arrival and avoid repeated freeze-thaw cycles to maintain performance.
