Biological Background: Neuronal Acetylcholine Receptor Subunit Alpha-4 (CHRNA4/ACHA4) Function and Localization
- Core component of pentameric neuronal nicotinic acetylcholine receptors (nAChRs), which function as ligand-gated cation channels with high calcium permeability. Related references: PMID:22361591, PMID:27698419, PMID:29720657
- Encoded by the CHRNA4 gene (aliases: NACRA4, ACHA4, EBN, BFNC) and each nAChR subunit confers distinct channel properties including activation kinetics, pH sensitivity, and allosteric modulation.
- Primarily forms heteropentameric receptors with CHRNB2 or CHRNB4; CHRNA4-containing receptors represent the most abundant nAChR subtype in the central nervous system. Related references: PMID:16835356, PMID:22361591, PMID:27698419
- Exists in two major stoichiometries, (CHRNA4)3:(CHRNB2)2 and (CHRNA4)2:(CHRNB2)3, which differ in unitary conductance, calcium permeability, ACh sensitivity, and potentiation by divalent cations. Related references: PMID:27698419, PMID:29720657, PMID:38454578
- Modulates calcium-dependent signaling pathways and influences the release of neurotransmitters such as dopamine, glutamate, and GABA (by similarity).
- Localized primarily at the synaptic cell membrane and the plasma membrane of neurons.
- Classified under keywords including alternative splicing, disease variant (epilepsy), glycoprotein, ion channel, and receptor; genetic variants are linked to epilepsy.
Experimental Guidance and Technical Tips
- The immunogen (aa 29-242 of human ACHA4) targets the N-terminal extracellular domain, allowing recognition of both (α4)3(β2)2 and (α4)2(β2)3 receptor stoichiometries.
- For Western blotting, a specific band at approximately 70 kDa is expected; reducing conditions and membrane protein extraction methods are recommended.
- In immunofluorescence/ICC, positive staining is anticipated at the plasma membrane and synaptic sites in neuronal cultures or cell lines expressing CHRNA4.
- For ELISA applications, the polyclonal nature may allow compatibility with various assay formats; confirm pairing with detection antibodies in your system.
- Cross-reactivity with mouse is reported; validate performance in mouse brain tissue or primary neuronal cultures.
