Biological Background: Neurogenic differentiation factor 1 Function and Localization
- Neurogenic differentiation factor 1 (NEUROD1), also known as Class A basic helix-loop-helix protein 3 (bHLHa3), is a member of the basic helix-loop-helix (bHLH) transcription factor family.
- Acts as a transcriptional activator by binding to E box-containing promoter consensus core sequences (5’-CANNTG-3’), and associates with the p300/CBP coactivator complex to stimulate expression of target genes such as secretin and the cyclin-dependent kinase inhibitor CDKN1A.
- Plays critical roles in neuronal and endocrine cell differentiation, contributing to the formation of early retinal ganglion cells, inner ear sensory neurons, cerebellar and hippocampal granule cells, pancreatic endocrine islet cells, and enteroendocrine cells of the small intestine.
- Together with PAX6 or SIX3, NEUROD1 is required for amacrine cell fate specification in the retina, and is also involved in dendrite morphogenesis and maintenance in the cerebellar cortex.
- NEUROD1 associates with chromatin at enhancer regulatory elements of genes encoding key transcriptional regulators of neurogenesis, thereby influencing neurodevelopmental gene expression programs.
- The protein localizes to both the cytoplasm and nucleus, consistent with its roles in signal-responsive nuclear translocation and transcriptional regulation.
- Classified as a phosphoprotein, and mutations in the NEUROD1 gene are associated with diabetes mellitus (MODY6), underscoring its importance in pancreatic beta-cell function and developmental biology.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 160-356 of human NEUROD1 (NP_002491.2), encompassing the conserved bHLH domain and C-terminal region; this region is highly similar between human and rat, supporting the observed cross-reactivity.
- For Western blot (WB) analysis, an expected band near 40 kDa can be detected; consider using nuclear or whole-cell extracts from neuronal or pancreatic beta-cell lines, and validate with appropriate positive and negative controls.
- For ELISA, the antibody may be suitable for direct or capture formats; coating with recombinant NEUROD1 protein or specific peptide antigens is recommended, with optimization of antibody concentration and blocking conditions.
