Biological Background: NEUROD1 Function and Localization
- NEUROD1 (Neurogenic differentiation factor 1), also known as BETA2, BHF-1, or bHLHa3, is a class A basic helix-loop-helix transcription factor encoded by the NEUROD1 gene (Gene ID: 4760).
- It functions as a transcriptional activator by binding to E box-containing consensus sequences (5'-CANNTG-3') in target gene promoters, associating with coactivator complexes such as p300/CBP to drive transcription of genes like secretin and CDKN1A.
- NEUROD1 is pivotal in neurogenesis, directing differentiation of early retinal ganglion cells, inner ear sensory neurons, cerebellar and hippocampal granule cells, and amacrine cells (in cooperation with PAX6 or SIX3).
- Beyond the nervous system, it is essential for endocrine cell development, including pancreatic islet β-cells and enteroendocrine cells of the small intestine, linking it to pathways of diabetes mellitus (MODY6).
- The protein localizes to both the cytoplasm and nucleus, consistent with its regulated nuclear import for transcriptional activity.
- Post-translational modifications include phosphorylation, and the protein is implicated in dendrite morphogenesis and maintenance in the cerebellar cortex.
- Keywords from UniProt: Activator, Cytoplasm, Nucleus, Developmental protein, Differentiation, DNA-binding, Neurogenesis, Phosphoprotein, Transcription regulation, and Disease variant (Diabetes mellitus).
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering the full-length human NEUROD1 (aa 1-356). This design may generate polyclonal antibodies recognizing multiple epitopes, offering robust detection in IHC-P and ELISA.
- For IHC-P, consider optimizing antigen retrieval and antibody dilution in relevant tissues (e.g., brain, pancreas). Expect nuclear staining with possible cytoplasmic background, and validate with appropriate controls.
- In ELISA, the antibody can be used as a capture or detection reagent; titration and cross-reactivity testing against related bHLH family members is recommended.
- Given the cross-reactivity with mouse, this antibody is suitable for studies in mouse models; however, independent validation in your experimental system is advised.
