Biological Background: Neuraminidase/NEU1 Function and Localization

  • Encoded by the NEU1 gene, sialidase-1 (neuraminidase-1) is a 45 kDa lysosomal exo-α-sialidase that catalyzes the removal of terminal sialic acid residues from glycoproteins and glycolipids. Its enzymatic activity requires assembly into a multienzyme complex. Related references: PMID:14695530, PMID:25153125, PMID:37205763
  • Sialidase-1 exhibits substrate preference for α2-3 and α2-6 sialyl linkages, contributing to glycoconjugate catabolism and lipid degradation.
  • NEU1 localizes predominantly to the lysosomal lumen and membrane; it can also traffic to the cell membrane.
  • Expression is highest in pancreas, followed by skeletal muscle, kidney, placenta, heart, lung, and liver; weak in brain.
  • Post-translational modifications include disulfide bond formation, N-linked glycosylation, and phosphorylation.
  • Mutations in NEU1 cause sialidosis, a lysosomal storage disorder characterized by lysosomal accumulation of sialylated substrates.

Experimental Guidance and Technical Tips

  • The immunogen spans the core catalytic domain (aa 48-415) of human NEU1 and may recognize both precursor and mature protein forms.
  • For Western blot, a predominant band near 45 kDa is expected in human and mouse lysates; consider including reducing agents in sample buffer.
  • In immunofluorescence, a punctate perinuclear pattern consistent with lysosomes is anticipated; co-staining with lysosomal markers is advised.
  • ELISA utility should be empirically validated using recombinant NEU1 protein or appropriate cell/tissue extracts as coating material.
