Biological Background: NEU3 Function and Localization
- NEU3, also known as sialidase-3 or ganglioside sialidase, is encoded by the NEU3 gene and belongs to the neuraminidase family of exo-alpha-sialidases.
- It catalyzes the removal of terminal sialic acid residues from gangliosides, glycoproteins, and oligosaccharides, with high catalytic efficiency for gangliosides such as GD1a, GM3, and GD3. Related references: PMID:10405317 PMID:10861246 PMID:11298736
- NEU3 modulates transmembrane signaling by altering ganglioside composition in the lipid bilayer and by directly interacting with receptors like EGFR, leading to desialylation and activation of downstream signaling in proliferating cells. Related references: PMID:17334392 PMID:25922362
- The protein contributes to clathrin-mediated endocytosis by regulating the sorting of endocytosed receptors to early and recycling endosomes. Related references: PMID:26251452
- Subcellularly, NEU3 localizes to the plasma membrane, caveolae, and membranes of endosomes (early and recycling) and lysosomes.
