Biological Background: Nesprin-3 Function and Localization
- Nesprin-3, also known as KASH domain-containing protein 3 (KASH3) or nuclear envelope spectrin repeat protein 3, is encoded by the SYNE3 gene (C14orf139, C14orf49, LINC00341) and is a component of the LINC (LInker of Nucleoskeleton and Cytoskeleton) complex.
- It functions as an anchoring protein that tethers the nucleus to the cytoskeleton by binding to plectin (PLEC), which interacts with the intermediate filament system, thereby mediating the connection between the nuclear lamina and the cytoskeleton.
- The LINC complex is essential for transmitting mechanical forces across the nuclear envelope, and Nesprin-3 plays a role in nuclear movement and positioning.
- In aortic endothelial cells, Nesprin-3 is required for flow-induced centrosome polarization and directional migration, and it contributes to the regulation of cell morphology.
- Nesprin-3 localizes to the outer nuclear membrane, nuclear envelope, and rough endoplasmic reticulum.
- It is expressed at the protein level in aortic endothelial cells.
- The protein has a molecular weight of approximately 112 kDa, consistent with the UniProt annotation, and features spectrin repeats, a coiled coil domain, and a transmembrane helix.
Experimental Guidance and Technical Tips
- The antibody was generated using a recombinant fusion protein corresponding to amino acids 750–935 of human Nesprin-3, a region within the spectrin repeat domain. This design may enable recognition of full-length Nesprin-3 and potential isoforms generated by alternative splicing.
- For Western blot (WB), human aortic endothelial cell lysates or other Nesprin-3-expressing cell lines are suitable positive controls. The expected band size is ~112 kDa. Consider using appropriate loading controls and validate reactivity in mouse samples if needed.
- For ELISA, the antibody may be used as a capture or detection reagent. Optimize coating and detection conditions using purified recombinant Nesprin-3 protein or cell extracts. Cross-reactivity with other KASH domain proteins should be evaluated in the experimental context.
- Since shipping is on ice bags, ensure proper storage upon arrival (e.g., aliquot and store at -20°C or -80°C as recommended) to maintain antibody stability.
