- Nesprin-1 (also known as SYNE1, Enaptin, MYNE1, KASH1) is a multi-isomeric modular protein that forms a linking network between organelles and the actin cytoskeleton to maintain subcellular spatial organization.
- As a core component of the LINC (Linker of Nucleoskeleton and Cytoskeleton) complex, it connects the nuclear lamina to the cytoskeleton, mediating mechanical force transmission across the nuclear envelope and regulating nuclear movement and positioning.
- Together with SUN1/2 and cytoplasmic dynein-dynactin motor complexes, Nesprin-1 may participate in nucleus-centrosome attachment and nuclear migration in neural progenitors, with SYNE1 and SYNE2 acting redundantly.
- It is required for centrosome migration to the apical cell surface during early ciliogenesis and may contribute to nuclear remodeling during sperm head formation via a probable SUN3:SYNE1/KASH1 LINC complex.
- Subcellular localization includes the nuclear outer membrane, nuclear envelope, cytoplasm, cytoskeleton, myofibril sarcomere, and Golgi apparatus.
- Widely expressed with high levels in skeletal and smooth muscles, heart, spleen, peripheral blood leukocytes, pancreas, cerebellum, stomach, kidney, and placenta; isoform GSRP-56 is predominantly expressed in heart and skeletal muscle.

- For western blotting, Nesprin-1 exists as multiple isoforms; the major isoform migrates at approximately 1011 kDa, while a smaller isoform may appear around 112 kDa. Use appropriate gel percentage (e.g., 4-6% SDS-PAGE) and extended transfer times to ensure high-molecular-weight proteins are efficiently transferred.
- For immunofluorescence/immunocytochemistry, anticipate staining at the nuclear envelope, cytoplasm, and possibly cytoskeletal structures. Validate with known subcellular markers.
- For ELISA, ensure the antigen coating and antibody dilution are optimized using the recombinant immunogen as a reference standard.
