Biological Background: Nephrin Function and Localization
- Nephrin (encoded by Nphs1) is a transmembrane adhesion receptor of the immunoglobulin superfamily, with multiple isoforms generated by alternative splicing; isoform 1 is the predominant form in adult kidney.
- It plays a critical role in the development and function of the kidney glomerular filtration barrier, regulating vascular permeability and anchoring the podocyte slit diaphragm to the actin cytoskeleton.
- Nephrin is also involved in skeletal muscle formation by regulating myoblast fusion, and is expressed in muscle during embryonic development and following acute injury.
- The protein is localized at the cell membrane, consistent with its single-pass transmembrane structure.
- Highly expressed in kidney glomerular podocytes; additionally detected in developing cerebellum, hindbrain, spinal cord, retina, hypothalamus, and skeletal muscle during myoblast fusion. Isoforms 1 and 2 are expressed in newborn brain and developing cerebellum.
- Nephrin undergoes N-linked glycosylation, contains disulfide bonds, and is phosphorylated; it is classified as a developmental protein with immunoglobulin-like repeats.
Experimental Guidance and Technical Tips
- This antibody was raised against a recombinant protein corresponding to the C-terminal cytoplasmic region (aa 1033-1256) of mouse Nephrin, recognizing an intracellular epitope.
- For immunofluorescence on paraffin sections (IF-P) or immunohistochemistry (IHC-P), consider including a permeabilization step to allow access to the intracellular domain, and validate in appropriate mouse tissue samples.
- In western blotting (WB), Nephrin is detected as a band of approximately 135–136 kDa in mouse kidney or skeletal muscle lysates; ensure proper sample preparation to preserve membrane protein integrity.
- As a rabbit monoclonal antibody, it offers high batch-to-batch consistency and is suitable for quantitative ELISA applications.
- Cross-reactivity with other species has not been tested; for non-mouse experiments, conduct species verification if needed.
