Biological Background: NELFE Function and Localization
- NELFE (Negative Elongation Factor E), also known as RD, RDBP, or RDP, is a 43 kDa RNA-binding protein that is an essential subunit of the human Negative Elongation Factor (NELF) complex.
- As part of the NELF complex, NELFE cooperates with DRB Sensitivity-Inducing Factor (DSIF) to induce transcriptional pausing of RNA Polymerase II, a key regulatory step in early elongation. The P-TEFb kinase complex subsequently phosphorylates the complex to release pausing. Related references: PMID:10199401 PMID:27256882 PMID:11940650 PMID:12612062
- NELFE provides the strongest RNA-binding activity of the NELF complex, likely mediating initial recruitment to nascent RNA transcripts. Related references: PMID:18303858 PMID:27256882 PMID:27282391
- The protein is localized in the nucleus and associates with chromosomes.
- NELFE is broadly expressed across human tissues, including heart, brain, lung, placenta, liver, skeletal muscle, kidney, and pancreas.
- Apart from its canonical role in transcription regulation, the NELF complex has been implicated in HIV-1 latency maintenance.
- The UniProt record annotates multiple post-translational modifications on NELFE, such as ADP-ribosylation, phosphorylation, and ubiquitination, along with structural features including coiled-coil domains and repetitive sequence elements.
Experimental Guidance and Technical Tips
- The immunogen was generated against the N-terminal region (aa 1–280). Consider epitope accessibility in native protein conformations for IHC-P experiments.
- Standard blocking and antibody dilution protocols for polyclonal antibodies should be optimized empirically for each application (WB, IHC-P, ELISA) and sample type.
- For western blotting, a band around 43 kDa is expected; validate specificity using appropriate positive and negative controls (e.g., knockout cell lines if available).
- In immunohistochemistry, antigen retrieval methods may be required; test both heat-induced and enzymatic retrieval to determine optimal staining conditions.
