Biological Background: NEK2 Function and Localization
- NEK2 is a serine/threonine-protein kinase, also known as HSPK 21, NimA-related protein kinase 2 (NimA-related protein kinase 2), and NimA-like protein kinase 1.
- It plays a critical role in centrosome separation and bipolar spindle formation during mitosis, ensuring high-fidelity chromosome segregation.
- NEK2 regulates kinetochore-microtubule attachment stability by phosphorylating NDC80, and controls the mitotic checkpoint via phosphorylation of CDC20 and MAD2L1.
- It phosphorylates centrosomal proteins such as CROCC, CEP250, NINL, CEP68, and CNTLN to promote their displacement from centrosomes, a process essential for centrosome disjunction. Related references: PMID:24554434, PMID:25704143, PMID:26220856
- NEK2 also phosphorylates CCDC102B, causing its dissociation from the centrosome and facilitating separation. Related references: PMID:30404835
- The protein is localized to the centrosome, spindle poles, kinetochores, and nucleus/nucleolus, reflecting its dynamic role throughout the cell cycle.
- Two major isoforms exist: isoform 1 and 2 are widely expressed, with isoform 4 found in testis; NEK2 is upregulated in various cancers.
- Post-translational modifications include phosphorylation, and the protein contains an ATP-binding domain and a coiled-coil region.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1-100), which may detect multiple isoforms if the sequence is conserved.
- For Western blot, a band at approximately 52kDa is expected; optimal antibody dilution should be determined empirically.
- When using this antibody for ELISA, consider pairing it with a matched detection antibody for sandwich assays; validate performance in the relevant sample system.
- Cross-reactivity with mouse and rat NEK2 has been indicated; verify sequence homology between species to inform experimental design.
