- NEIL1 (Endonuclease 8-like 1), also known as DNA glycosylase/AP lyase Neil1, Nei homolog 1 (NEH1), and Nei-like protein 1, is a multifunctional DNA repair enzyme.
- It plays a critical role in base excision repair of DNA damaged by oxidation or mutagenic agents, acting as a DNA glycosylase that recognizes and removes damaged bases with a preference for oxidized pyrimidines (e.g., thymine glycol, formamidopyrimidine, 5-hydroxyuracil) and marginal activity toward 8-oxoguanine.
- Possesses AP lyase activity, cleaving the DNA backbone via beta-delta elimination to generate a single-strand break with both 3'- and 5'-phosphates.
- Exhibits DNA glycosylase/lyase activity toward mismatched uracil and thymine, particularly in U:C and T:C contexts, and specifically binds 5-hydroxymethylcytosine (5hmC), suggesting a reader function.
- Localizes to multiple subcellular compartments, including the cytoplasm (cytoskeleton, microtubule organizing center, centrosome), nucleus, and chromosome.
- Ubiquitously expressed in human tissues; belongs to the FPG/Nei family of DNA glycosylases.

- The immunogen covers the N‑terminal region (amino acids 1–135) of human NEIL1. Verify sequence homology across species of interest, as antibody reactivity may depend on this region.
- For Western blotting, a band corresponding to the calculated molecular weight of approximately 43.7 kDa (observed ~44 kDa) is expected. Include appropriate positive control lysates from human, mouse, or rat cells.
