Biological Background: NEDD9 Function and Localization
- NEDD9, also known as HEF1, CAS-L, and CASS2, is a scaffolding protein belonging to the Cas family, characterized by multiple protein interaction domains (SH3 domain) and a C-terminal domain involved in focal adhesion targeting.
- Functions as a central coordinator of tyrosine kinase signaling at focal adhesions, regulating cell adhesion, migration, and cytoskeletal reorganization. Related references: PMID:24574519
- Mediates integrin β1 and B-cell receptor signaling in lymphocytes, promoting lymphocyte adhesion and migration, and is required for proper lymphocyte homing to secondary lymphoid organs. Related references: PMID:9020138 PMID:17174122
- Plays a role in T-cell immunological synapse formation by organizing the cortical actin cytoskeleton and centralizing TCR microclusters.

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 390–560, located in the central region of NEDD9; epitopes may be preserved across human, mouse, and rat due to high homology.
- For Western blotting, expect a band around 93 kDa; however, alternative splicing and post-translational modifications may result in multiple bands.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), fixation and antigen retrieval conditions should be optimized; nuclear and cytoplasmic localization may be observed.
- For ELISA, the recombinant protein used as immunogen can serve as a positive control; note that the polyclonal nature may cause batch-to-batch variability.
- Cross-adsorption may be considered if cross-reactivity with other Cas family proteins (BCAR1, EFS) needs to be minimized.
