Biological Background: NDUFV1 Function and Localization
- Encodes NADH dehydrogenase [ubiquinone] flavoprotein 1, also known as CI-51kD, UQOR1, or NDUFV1, a core subunit of mitochondrial Complex I.
- Functions as the initial electron acceptor in Complex I, oxidizing NADH and transferring electrons via FMN and iron-sulfur clusters to ubiquinone. Related references: PMID:28844695
- Contains one FMN cofactor and one 4Fe-4S iron-sulfur cluster, essential for electron tunnelling within the peripheral arm of the enzyme.
- Localized to the mitochondrial inner membrane, where it participates in the proton-translocating NADH-ubiquinone oxidoreductase activity.
- Subject to post-translational modifications including acetylation and methylation; mutations are associated with Leigh syndrome (MC1DN4).
- Alternative splicing generates isoforms that may contribute to functional diversity or regulation of Complex I assembly.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 72-420, covering a large portion of the mature protein; this region is conserved across human, mouse, and rat, consistent with the observed cross-reactivity.
- For Western blot (WB), a band near 51 kDa is expected; consider using mitochondrial-enriched lysates to enhance signal.
- In immunohistochemistry (IHC-P), optimize antigen retrieval and antibody dilution; include appropriate tissue controls (e.g., heart, skeletal muscle, brain) known to have high mitochondrial content.
- For ELISA, pair with a suitable detection system and validate linearity and sensitivity in your sample matrix.
- As a polyclonal antibody raised in rabbit, lot-to-lot consistency should be verified; consider using a blocking buffer compatible with rabbit primary antibodies.
