Biological Background: NDUFS2 Function and Localization
- NDUFS2 encodes the NADH dehydrogenase [ubiquinone] iron-sulfur protein 2, also known as Complex I-49kD (CI-49kD) and NADH-ubiquinone oxidoreductase 49 kDa subunit. It is a core subunit of mitochondrial respiratory chain Complex I.
- Essential for the catalytic activity of Complex I, which catalyzes electron transfer from NADH to ubiquinone. Related references: PMID:22036843, PMID:28031252, PMID:30922174
- Plays a critical role in the assembly of Complex I (By similarity). It is also a redox-sensitive component of the oxygen-sensing pathway in the pulmonary vasculature, mediating acute hypoxic pulmonary vasoconstriction. Related references: PMID:30922174
- Contributes to glia-like neural stem and progenitor cell proliferation, differentiation, and subsequent oligodendrocyte or neuronal maturation (By similarity).
- Subcellular localization: mitochondrial inner membrane.
- Keywords associated include iron-sulfur cluster binding (4Fe-4S), electron transport, oxidoreductase activity, and involvement in primary mitochondrial disease. Mutations in NDUFS2 are associated with Leber hereditary optic neuropathy and mitochondrial complex I deficiency.
Experimental Guidance and Technical Tips
- For Western blotting, consider using mitochondrial-enriched fractions from human, mouse, or rat tissues to detect endogenous NDUFS2 at approximately 53 kDa (apparent molecular weight may slightly differ from the calculated 52.5 kDa due to post-translational modifications).
- Validate antibody specificity by including appropriate positive and negative controls (e.g., NDUFS2 knockout or siRNA-mediated knockdown samples) in your experimental setup.
- For ELISA, the antibody may be screened as a capture or detection reagent after pairing optimization with another NDUFS2-specific antibody or recombinant protein standards. Assess linearity and sensitivity in relevant biological matrices.
- The immunogen (amino acids 300-434) targets a region likely exposed in native or partially denatured conditions, supporting detection of both native and denatured protein; confirm performance under your specific assay conditions.
