Biological Background: NDUFAF4 Function and Localization
- NDUFAF4 (NADH dehydrogenase [ubiquinone] 1 alpha subcomplex assembly factor 4) is also known as hormone-regulated proliferation-associated protein of 20 kDa (HRPAP20). It belongs to the complex I NDUFAF4 assembly factor family.
- Plays a critical role in the assembly of mitochondrial NADH:ubiquinone oxidoreductase (complex I), a key component of the oxidative phosphorylation system (PubMed:18179882, PubMed:28853723). Related references: PMID:18179882 PMID:28853723
- May be involved in cell proliferation and survival of hormone-dependent tumor cells, and has been suggested as a regulator of breast tumor cell invasion.
- Localizes to the mitochondrion membrane, consistent with its role in complex I assembly.
- Post-translational modifications include myristoylation and phosphorylation, and it is predicted to be a lipoprotein.
- Contains a calmodulin-binding domain and is associated with host-virus interactions.
- Mutations in NDUFAF4 are linked to primary mitochondrial disease (MC1DN15).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length human NDUFAF4 protein (amino acids 1–170). Consider using it to detect both endogenous and overexpressed NDUFAF4 in relevant sample types.
- For Western blot, a band at ~20 kDa is expected. Validate the antibody in whole-cell lysates from human, mouse, or rat cell lines expressing NDUFAF4.
- In immunohistochemistry (IHC-P), antigen retrieval may be required. Titrate the antibody to achieve optimal signal-to-noise ratio in mitochondrial-rich tissues.
- For immunofluorescence (IF/ICC), co-staining with a mitochondrial marker is recommended to confirm subcellular localization.
- This polyclonal antibody is suitable for ELISA; however, optimal coating and detection conditions should be determined experimentally.
