  • Protein NDRG2, also designated N-myc downstream-regulated gene 2 protein or Syld709613, is implicated in tumor suppression and developmental processes.
  • It contributes to the regulation of the Wnt signaling pathway by down-regulating CTNNB1-mediated transcriptional activation of target genes such as CCND1, suggesting a tumor suppressor role.
  • NDRG2 may also be involved in dendritic cell differentiation and neuron differentiation.
  • Subcellularly, the protein localizes to the cytoplasm, with enrichment in the perinuclear region, and is found in cell projection growth cones.
  • Tissue expression is highest in brain, heart, skeletal muscle, and salivary gland, with moderate levels in kidney and liver. It is expressed in dendritic cells but not in other blood cells. Expression is low or absent in several cancer types, including pancreatic, liver, and glioblastoma.
  • Two major isoforms (isoform 1 and isoform 2) are present in brain neurons and are up-regulated in Alzheimer disease.
  • Post-translational modifications include acetylation and phosphorylation, and the protein undergoes alternative splicing, highlighting regulatory complexity.
