Biological Background: NDP52 Function and Localization
- NDP52, also known as Calcium-binding and coiled-coil domain-containing protein 2 (CALCOCO2), Antigen nuclear dot 52 kDa protein, and Nuclear domain 10 protein NDP52, is a 52 kDa protein encoded by the CALCOCO2 gene.
- Functions as a xenophagy-specific receptor: targets cytosolic bacteria such as Salmonella typhimurium to autophagosomes via interactions with MAP1LC3A/B and GABARAPL2, and regulates autophagosome maturation through MAP3LC3C, thereby restricting pathogen proliferation. - May also play a role in ruffle formation, actin cytoskeleton organization, and negative regulation of constitutive secretion. Related references: PMID:17635994
- Localizes to the cytoplasm, particularly the perinuclear region, and associates with the cytoskeleton and autophagosome membranes.
- Expressed in all tissues tested, with highest levels in skeletal muscle and lowest in brain.
