Biological Background: NDEL1 (NUDEL) Function and Localization
- Encoded by the NDEL1 gene, this protein is also known as Nuclear distribution protein nudE-like 1 (NUDEL) or Mitosin-associated protein 1 (MITAP1). It is a key regulator of microtubule organization and dynein-mediated processes.
- Required for organizing the cellular microtubule array and anchoring microtubules at the centrosome, partly by targeting the microtubule-severing protein KATNA1 to the centrosome.
- Positively regulates dynein motor activity by targeting dynein to microtubule plus ends, enabling dynein-dependent functions such as maintenance of Golgi integrity, centripetal motion of secretory vesicles, and coupling of nucleus and centrosome.
- Essential for neuronal migration during brain development: guides newly formed neurons from the ventricular/subventricular zone to the cortical plate, and together with DISC1, regulates neurite outgrowth.
- May act as a RAB9A/B effector that tethers RAB9-associated late endosomes to the dynein motor for retrograde transport to the trans-Golgi network. Related references: PMID:34793709
- Facilitates polymerization of neurofilaments from NEFH and NEFL subunits and positively regulates lysosome peripheral distribution in osteoclasts (by similarity).
- Subcellularly localizes to the cytoplasm, cytoskeleton, centrosome, kinetochore, and mitotic spindle. Widely expressed in brain, heart, kidney, liver, lung, pancreas, placenta, and skeletal muscle. Features a coiled-coil domain and is subject to palmitoylation and phosphorylation.

- The immunogen corresponds to amino acids 1-345 of human NDEL1 (NP_110435.1), covering nearly the full-length protein. This broad immunogen design may contribute to reliable detection in both denaturing western blot (WB) and native ELISA applications.
- For western blotting, the predicted molecular weight is approximately 38 kDa; consider loading appropriate protein ladders and validating band size in relevant human or mouse cell/tissue lysates.
- Given reported human and mouse cross-reactivity, this antibody is suitable for comparative studies across these species. We recommend initial titration and optimization of antibody dilution for each application and sample type.
