Biological Background: NDE1 Function and Localization

  • Nuclear distribution protein nudE homolog 1 (NDE1, also known as NUDEL) is a key regulator of cell division and neurogenesis. It belongs to the nudE family of microtubule-associated proteins.
  • NDE1 is essential for centrosome duplication and proper mitotic spindle formation, ensuring accurate chromosome segregation during mitosis.
  • In the developing cerebral cortex, NDE1 controls the orientation of the mitotic spindle in neuronal progenitors; a perpendicular division yields two progenitors, while a parallel division produces one progenitor and one neuron, thus regulating the balance between proliferation and differentiation.
  • As a RAB9A/B effector, NDE1 tethers RAB9-associated late endosomes to the dynein motor complex, facilitating their retrograde transport to the trans-Golgi network.

  • Subcellularly, NDE1 localizes to the cytoplasm, centrosome, spindle apparatus, kinetochore, cleavage furrow, and cytoplasmic vesicle membranes, consistent with its roles in mitosis and endosomal trafficking.

  • The immunogen used is a recombinant fusion protein spanning amino acids 1–110 of human NDE1, which covers the N-terminal region; this region is conserved across human, mouse, and rat, consistent with the predicted cross-reactivity.
  • For Western blot (WB), a band around 38 kDa (matching the predicted molecular weight) is expected. We recommend titrating the antibody to determine the optimal dilution for your specific sample and detection system.
  • In immunohistochemistry (IHC-P), antigen retrieval and blocking steps should be optimized to achieve strong, specific staining. Use of appropriate tissue controls (e.g., brain or cortex sections) is recommended.
  • For ELISA, the polyclonal nature may provide high sensitivity; consider using the antibody in a capture or detection format and confirm specificity with appropriate controls.
