Biological Background: NCTR2 Function and Localization
- NCTR2, also known as NKp44, CD336, or LY95, is a natural cytotoxicity triggering receptor encoded by the NCR2 gene.
- It functions as a cytotoxicity-activating receptor that enhances the ability of activated natural killer (NK) cells to lyse tumor cells.
- The protein is a single-pass type I membrane protein located on the cell membrane (UniProt Keywords: Cell membrane, Transmembrane, Transmembrane helix).
- Selectively expressed on activated NK cells and in vitro cultured TCRγδ lymphoid cells, underlining its role in immune surveillance.
- Contains an immunoglobulin domain, a signal peptide, and disulfide bonds (UniProt Keywords: Immunoglobulin domain, Signal, Disulfide bond, Glycoprotein).
- Undergoes alternative splicing (UniProt Keywords: Alternative splicing) and is subject to post-translational modifications including glycosylation and disulfide bond formation.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal region (aa 1-100) of human NCTR2, which includes the signal peptide and part of the extracellular domain. This design may favor detection of the mature protein.
- For Western blot, consider that NCTR2 has a predicted molecular weight of ~31 kDa (UniProt: 30677 Da), but glycosylation and signal peptide cleavage may result in a band around 31 kDa; validate with appropriate controls.
- For ELISA, the antibody may be used for direct detection or as a capture antibody; consider pairing with a different clone for sandwich ELISA.
- Since expression is restricted to activated NK cells and TCRγδ cells, use appropriate cell lysates or stimulated samples to confirm reactivity in WB.
