Biological Background: NCOA7 Function and Localization

  • NCOA7 (Nuclear receptor coactivator 7) is also known as ERAP140, ESNA1, and TLDC4. It enhances transcriptional activities of nuclear receptors including ESR1, THRB, PPARG, and RARA. Related references: PMID:11971969
  • The protein inhibits vacuolar-type ATPase (V-ATPase) by inducing disassembly of the complex, protecting the Golgi lumen from excess acidification and maintaining glycosylation enzyme conditions. Related references: PMID:38593795
  • NCOA7 is involved in protection against oxidative damage and exhibits oxidation resistance activity, protecting neuronal cells from oxidative stress. Related references: PMID:17391516, PMID:25330068
  • Isoforms with stronger neuroprotective activity also inhibit endosome-mediated viral entry by blocking viral-host membrane fusion and promoting lysosomal degradation of endocytosed material. Related references: PMID:30478388
  • The protein localizes to the nucleus, cytoplasm, and Golgi apparatus (cis- and trans-Golgi network membranes), consistent with roles in transcription and membrane trafficking.
  • It is highly expressed in brain, with weaker expression in mammary gland, ovary, uterus, prostate, stomach, bladder, spinal cord, pancreas, and cancer cell lines.
  • Post-translational modifications include acetylation and phosphorylation; the protein features coiled-coil domains.
  • The gene undergoes alternative splicing, producing isoforms with molecular weights ranging from 12 kDa to 106 kDa.

Experimental Guidance and Technical Tips

  • The immunogen spans a region common to several NCOA7 isoforms; consider verifying reactivity with the isoform of interest in your experimental system.
  • For Western blot, multiple bands between 12 kDa and 106 kDa may be observed due to alternative splicing; use appropriate positive controls such as brain or cancer cell lines.
  • For ELISA, validate with recombinant protein or cell lysates; cross-reactivity with mouse allows studies in murine models.
  • Optimize antibody dilution for each application and sample type; preliminary titration is recommended.
