Biological Background: NCKAP1 Function and Localization
- Nck-associated protein 1 (NCKAP1), also designated HEM2, NAP1, NAP125, or p125Nap1, is a 128.8 kDa protein encoded by the NCKAP1 gene in humans.
- NCKAP1 is a core component of the WAVE regulatory complex, which promotes lamellipodia formation by regulating actin filament reorganization through the Arp2/3 complex, a process activated by the small GTPase RAC1.
- Within the WAVE1 complex, NCKAP1 is required for brain-derived neurotrophic factor (BDNF)-induced endocytic trafficking and signaling from early endosomes via the NTRK2 receptor.
- The protein localizes to the cell membrane and is enriched at the lamellipodium membrane, consistent with its role in cell motility.
- NCKAP1 is widely expressed across human tissues, with highest levels in brain, heart, and skeletal muscle; it is notably absent in peripheral blood leukocytes.
