- NCF4/p40-phox is a subunit of the phagocyte NADPH oxidase complex that generates superoxide anions by transferring electrons from NADPH to molecular oxygen.
- The protein contains SH3 and PX domains, classifying it as an SH3 and PX domain-containing protein 4 (SH3PXD4); it is also known as neutrophil NADPH oxidase factor 4.
- Activation of the complex requires assembly of cytosolic subunits, including NCF4, with the membrane-bound core complex; this process is initiated by phosphorylation-dependent binding of NCF1/p47-phox to CYBA/p22-phox.
- NCF4 expression is restricted to hematopoietic cells.
