- NCBP2 (also known as CBP20, NIP1, CBC2, PIG55) is the 20 kDa subunit of the nuclear cap-binding complex (CBC), which binds co-transcriptionally to the 5' cap of pre-mRNAs and is essential for multiple aspects of RNA metabolism.
- As part of the CBC, NCBP2 participates in pre-mRNA splicing, mRNA export (including U snRNA and intronless mRNA export), pioneer round translation, nonsense-mediated mRNA decay (NMD), and microRNA biogenesis.
- The CBC complex is critical for the pioneer round of translation, during which NMD takes place; it is later replaced by eIF4E for steady-state translation.
- NCBP2 directly recognizes and binds the m7GpppG cap structure, but requires NCBP1/CBP80 to stabilize the high-affinity cap-binding conformation.
