- NCAM1 (Neural cell adhesion molecule 1), also known as CD56 and MSK39, is a glycoprotein of the immunoglobulin superfamily that mediates cell–cell adhesion through homophilic and heterophilic interactions.
- It plays a critical role in nervous system development, including neuron-neuron adhesion, neurite fasciculation, and promotion of neurite outgrowth.
- NCAM1 is expressed as multiple isoforms via alternative splicing, including transmembrane and glycosylphosphatidylinositol (GPI)-anchored variants, with a calculated molecular mass of approximately 94.6 kDa (mature protein ~95–120 kDa due to glycosylation).
- Subcellularly, NCAM1 localizes predominantly to the cell membrane as a single-pass type I membrane protein, but secreted and GPI-anchored forms also exist.
- Post-translational modifications include N-linked glycosylation, disulfide bond formation, and phosphorylation on cytoplasmic residues; the GPI-anchored isoform is lipidated.
- NCAM1 has been identified as a receptor for viral entry; it acts as a host cell receptor for rabies virus and Zika virus.
