Biological Background: NBS1/NBN Function and Localization
-
NBS1/NBN is a key component of the MRE11-RAD50-NBN (MRN) complex that plays a central role in DNA double-strand break (DSB) repair, homologous recombination (HR), telomere integrity maintenance, and meiosis. Related references: PMID:10888888, PMID:15616588, PMID:18411307
-
Within the MRN complex, NBN serves as a protein-protein adapter that specifically recognizes and binds phosphorylated proteins, mediating the recruitment of DNA repair factors and signaling kinases such as MRE11, RAD50, ATM, ATR, and DNA-PKcs to DSB sites. Related references: PMID:12419185, PMID:18411307, PMID:18583988, PMID:15064416, PMID:15790808
-
The MRN complex facilitates DNA end resection through MRE11 endonuclease and exonuclease activities, generating single-stranded DNA for HR; NBN additionally recruits phosphorylated CtIP (RBBP8) to promote resection and clear protein-DNA adducts. Related references: PMID:19759395, PMID:28867292, PMID:9705271, PMID:27814491, PMID:27889449, PMID:30787182
-
NBS1/NBN contributes to telomere length regulation by interacting with TERF2 during G1 phase to prevent DCLRE1B/Apollo recruitment, and CDK2-mediated phosphorylation of NBN dictates repair pathway choice (NHEJ vs. MMEJ) at dysfunctional telomeres. Related references: PMID:10888888, PMID:28216226
-
NBN is ubiquitously expressed with high levels in testis and localizes to the nucleus, chromosomes, PML bodies, and telomeres, consistent with its roles in DNA repair and cell cycle regulation.
Experimental Guidance and Technical Tips
- The antibody was raised against a synthetic peptide corresponding to amino acids 655-754 of human NBN (O60934). This region lies within the C-terminal domain, which mediates protein-protein interactions, and is expected to detect full-length NBN. For isoform-specific applications, consider using peptides mapping to other regions.
- Validated applications include Western blot (WB), immunoprecipitation (IP), and ELISA with human samples. For WB, NBN protein weight is ~85 kDa. Endogenous expression varies; testis and actively cycling cells are likely to show higher signals.
- For IP, optimize antibody concentration and lysis buffer conditions to preserve the MRN complex integrity, as NBN is tightly associated with MRE11 and RAD50.
- For ELISA, the synthetic peptide immunogen or recombinant NBN protein can serve as coating antigen. Include appropriate controls.
