Biological Background: NBAS Function and Localization

• NBAS (neuroblastoma-amplified sequence) is a subunit of the NRZ tethering complex that mediates Golgi-to-endoplasmic reticulum (ER) retrograde transport by facilitating SNARE assembly at the ER. Related references: PMID:19369418
• Essential for normal embryonic development.
• May play a role in the nonsense-mediated decay pathway of mRNAs containing premature stop codons.
• Localizes predominantly to the cytoplasm, ER, and ER membrane.
• Broadly expressed with highest levels in heart and skeletal muscle; low in liver, small intestine, and thymus. Up-regulated in some neuroblastoma cell lines.
• Contains WD repeat motifs and undergoes post-translational modifications including phosphorylation and acetylation.

Experimental Guidance and Technical Tips

• The immunogen spans the N-terminal region (aa 1–70). Consider using this antibody for detecting full-length NBAS or N-terminal fragments.
• NBAS has a predicted molecular weight of ~269 kDa. For Western blot, use a low-percentage (e.g., 6–8%) SDS-PAGE gel and wet transfer with extended time and low voltage to ensure efficient transfer of high-molecular-weight proteins.
• Validate antibody specificity in your target tissue or cell line; expression is widespread but levels vary, with highest abundance in heart and skeletal muscle.
• For immunofluorescence/ICC, optimize permeabilization conditions to detect ER membrane-associated NBAS.
• The antibody cross-reacts with human and rat samples; for other species, sequence homology should be checked before use.
