Biological Background: NAXE Function and Localization
- NAXE (NAD(P)H-hydrate epimerase), also known as AIBP (apolipoprotein A-I-binding protein), APOA1BP, YJEFN1, and PEBEL, is encoded by the NAXE gene. It belongs to the YjeF N-terminal domain-containing protein family.
- Functions as an epimerase that catalyzes the interconversion of S- and R-forms of NAD(P)HX, a damaged form of NAD(P)H resulting from enzymatic or heat-dependent hydration. This epimerization is a prerequisite for the S-specific NAD(P)H-hydrate dehydratase to repair both epimers, maintaining NAD(P)H cofactor integrity.

PMID:27616477 - In endothelial cells, NAXE accelerates cholesterol efflux to high-density lipoprotein (HDL), thereby regulating angiogenesis. This function links cellular lipid metabolism to vascular homeostasis.
Related references:
PMID:23719382 - The protein localizes predominantly to mitochondria but can also be secreted, as evidenced by its presence in cerebrospinal fluid, urine, and serum of sepsis patients.
- NAXE is ubiquitously expressed, with highest levels in kidney, heart, and liver. At the protein level, it is found in body fluids under certain pathological conditions.
- Post-translational modifications include phosphorylation, and the protein is annotated as a phosphoprotein. Alternative splicing produces multiple transcript variants.
- Keywords associated with NAXE include isomerase activity, lipid transport, metal-binding, nucleotide-binding, and its involvement in neurodegeneration, highlighting its multifaceted roles in metabolism and disease.
Experimental Guidance and Technical Tips
- The immunogen used to generate this antibody comprises amino acids 48–170 of human NAXE, a region conserved across human, mouse, and rat, supporting the observed cross-reactivity. For optimal results, verify cross-reactivity in your specific experimental setup.
- In Western blotting, the antibody detects NAXE with an estimated molecular weight of ~32 kDa (calculated from the human sequence: 31.7 kDa); however, always use appropriate molecular weight markers and validate bands in your lysate.
- For ELISA, the antibody can be employed to capture or detect NAXE. Given its secretion into body fluids in certain diseases, you may consider testing cerebrospinal fluid, urine, or serum (e.g., sepsis samples) as sample matrices, with proper controls.
