Biological Background: Nav1.5 Function and Localization

  • Nav1.5, encoded by the SCN5A gene, is the pore-forming alpha subunit of the cardiac voltage-gated sodium channel, also known as sodium channel protein cardiac muscle subunit alpha or hH1.
  • It directly mediates the depolarizing phase of action potentials by enabling selective Na+ influx, a process essential for membrane excitability and electrical signal propagation. Related references: PMID:33712541 PMID:40768348 PMID:1309946 PMID:21447824 PMID:23085483
  • As the predominant sodium channel in myocardial cells, Nav1.5 initiates the heartbeat by driving the upstroke of the cardiac action potential. Related references: PMID:11234013 PMID:11804990 PMID:12569159
  • Nav1.5 is required for normal electrical conduction, including formation of the infranodal ventricular conduction system, through interaction with XIRP2.
  • Subcellularly, Nav1.5 localizes to the sarcolemma, T-tubules, cell membrane, perinuclear cytoplasm, and cell junctions.
  • Tissue expression is predominantly in atrial and ventricular cardiac muscle, with additional detection in jejunal smooth muscle; isoform 4 is brain-specific.
  • Mutations in SCN5A are associated with Brugada syndrome, long QT syndrome type 3, atrial fibrillation, and cardiomyopathy; post-translational modifications include methylation, phosphorylation, ubiquitination, and glycosylation.

Experimental Guidance and Technical Tips

  • For Western blotting, anticipate multiple bands around 227 kDa, 223 kDa, 221 kDa, or 225 kDa due to glycosylation or isoform variations; validate detection using appropriate positive and negative controls.
  • In immunohistochemistry (paraffin-embedded) experiments, optimize antigen retrieval conditions and antibody dilution based on your specific tissue type; cardiac muscle sections are a recommended positive control.
  • This antibody can be used in ELISA for quantitative or qualitative detection of human Nav1.5; consider direct or indirect coating strategies and verify cross-reactivity with your sample matrix.
  • The immunogen corresponds to amino acids 273–357 of human Nav1.5; confirm that the epitope region is accessible in your assay format and target conformation.
