Biological Background: Nav1.5/SCN5A Function and Localization

  • Nav1.5 (SCN5A) is the pore-forming alpha subunit of the voltage-gated sodium channel Nav1.5, responsible for the depolarizing phase of action potentials in excitable membranes. Related references: PMID:33712541 PMID:40768348
  • Nav1.5 is the predominant sodium channel in myocardial cells, mediating the initial upstroke of the cardiac action potential and initiating the heartbeat. Related references: PMID:11234013 PMID:11804990 PMID:12569159
  • It allows selective Na+ ion influx upon membrane depolarization, propagating electrical signals throughout cardiac tissues. Related references: PMID:1309946 PMID:21447824 PMID:23085483
  • Required for normal electrical conduction and action potential configuration, partly through interaction with XIRP2 (By similarity).
  • Alternative splicing generates multiple isoforms; isoform 4 is expressed in the brain, while the full-length channel is cardiac-specific.
  • Localizes to the sarcolemma, T-tubules, and perinuclear cytoplasm in cardiomyocytes, ensuring rapid impulse transmission.
  • Mutations in SCN5A cause cardiac arrhythmia syndromes such as long QT syndrome type 3 (LQT3), Brugada syndrome, and atrial fibrillation.
  • Undergoes phosphorylation, methylation, ubiquitination, and glycosylation, which modulate channel trafficking and function.

Experimental Guidance and Technical Tips

  • For Western blotting (WB), consider using 6-8% SDS-PAGE gels and prolonged transfer times (e.g., 2-3 hours at 100 V) due to the high molecular weight of Nav1.5 (221-227 kDa). Include a high-range molecular weight marker for accurate size confirmation.
  • For immunohistochemistry (IHC-P), antigen retrieval with citrate buffer (pH 6.0) or EDTA (pH 9.0) may improve epitope exposure; validate with appropriate positive (cardiac muscle) and negative (e.g., liver) tissue controls.
  • The immunogen (aa 273-357) resides within an intracellular loop; thus, permeabilization steps (e.g., 0.1% Triton X-100) are recommended for immunofluorescence or staining protocols requiring access to cytoplasmic epitopes.
  • For ELISA, the antibody could be used as a capture or detection reagent for Nav1.5 in cell or tissue lysates; pairing with another clone or a polyclonal antibody may enhance sensitivity.
  • Since the antibody cross-reacts with mouse and rat Nav1.5, it is suitable for comparative studies across species; however, verify specificity in the species of interest using knockout cell lines or siRNA knockdown if possible.
