Biological Background: Nav1.5/SCN5A Function and Localization
- Nav1.5 (SCN5A) is the pore-forming alpha subunit of the voltage-gated sodium channel Nav1.5, responsible for the depolarizing phase of action potentials in excitable membranes. Related references: PMID:33712541 PMID:40768348

- For immunohistochemistry (IHC-P), antigen retrieval with citrate buffer (pH 6.0) or EDTA (pH 9.0) may improve epitope exposure; validate with appropriate positive (cardiac muscle) and negative (e.g., liver) tissue controls.
- The immunogen (aa 273-357) resides within an intracellular loop; thus, permeabilization steps (e.g., 0.1% Triton X-100) are recommended for immunofluorescence or staining protocols requiring access to cytoplasmic epitopes.
