Biological Background: NAT10 Function and Localization

  • NAT10 (N-acetyltransferase 10), also known as hALP, ALP, Kre33, and NET43, is an RNA cytidine acetyltransferase that catalyzes the formation of N4-acetylcytidine (ac4C) on a broad range of mRNAs, 18S rRNA, and tRNAs, thereby enhancing mRNA stability and translation. Related references: PMID:25411247 PMID:25653167 PMID:30449621 PMID:35679869
  • It specifically mediates ac4C modification at position 1842 in 18S rRNA (and possibly position 1337), and is required for early nucleolar cleavages at sites A0, A1, and A2 during 18S rRNA synthesis. Related references: PMID:25411247 PMID:25653167
  • In addition to its RNA acetyltransferase activity, NAT10 can acetylate protein lysine residues in vitro, including histones, microtubules, p53, and MDM2; however, the in vivo relevance of this activity remains uncertain. Related references: PMID:14592445 PMID:17631499 PMID:19303003 PMID:30449621
  • NAT10 activates telomerase by stimulating TERT gene transcription and may regulate telomerase function by affecting subunit assembly, disassembly, and localization. Related references: PMID:14592445 PMID:18082603
  • It is involved in centrosome duplication by acetylating CENATAC during mitosis, which promotes proteasomal degradation of SASS6. Related references: PMID:31722219
  • NAT10 is a component of the small subunit (SSU) processome, the earliest precursor of the small eukaryotic ribosomal subunit, where it participates in pre-rRNA folding, modification, and cleavage. Related references: PMID:34516797
  • The protein localizes predominantly to the nucleolus and is also found at the midbody during cell division, consistent with its roles in ribosome biogenesis and mitosis.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 750–1025 of human NAT10; when designing complementary antibodies or overlapping peptides for sandwich assays, consider targeting regions outside this fragment.
  • This rabbit monoclonal antibody has been validated for intracellular flow cytometry; ensure appropriate fixation and permeabilization steps for optimal detection of nuclear targets.
  • NAT10 is predominantly a nucleolar protein; for detection in other applications such as Western blot or immunohistochemistry, validate the antibody in the relevant sample system and consider nuclear enrichment or antigen retrieval protocols.
  • The antibody’s performance in other applications (e.g., WB, IP, IF) has not been assessed; if extending use, perform thorough validation with appropriate positive and negative controls.
