Biological Background: Nanog Function and Localization
- Homeobox protein NANOG is a transcription regulator critical for inner cell mass and embryonic stem (ES) cell proliferation and self-renewal.
- Imposes pluripotency on ES cells and prevents differentiation towards extraembryonic endoderm and trophectoderm lineages.
- Acts as both a transcriptional activator and repressor; binds to DNA consensus sequences 5'-TAAT[GT][GT]-3' or 5'-[CG][GA][CG]C[GC]ATTAN[GC]-3', and to the POU5F1/OCT4 promoter. Related references: PMID:25825768
- Blocks bone morphogenetic protein-induced mesoderm differentiation by physically interacting with SMAD1 and interfering with coactivator recruitment.
- Able to autorepress its own expression in differentiating ES cells via interaction with ZNF281/ZFP281, leading to NuRD complex recruitment and repression.
- Subcellular localization is exclusively nuclear, consistent with its role as a transcription factor.
- Expressed in testicular carcinoma and derived germ cell tumors, fetal gonads, and embryonic carcinoma cell lines; not expressed in many somatic organs.
- Keywords: alternative splicing, homeobox domain, developmental protein, DNA-binding, nuclear protein.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal region (aa 1–105), which may detect full-length and N-terminal isoforms; verify specificity in the relevant sample system.
- For WB, consider a predicted molecular weight of ~35 kDa (observed 35 kDa per supplier), but actual migration may vary; use appropriate positive and negative controls.
- For IHC-P, validate in tissue sections with known NANOG expression, such as embryonic carcinoma or testicular germ cell tumors; optimize antigen retrieval conditions.
- ELISA performance depends on coating strategy and detection format; consider using recombinant NANOG protein standards for assay calibration.
- As a polyclonal antibody, lot-to-lot consistency should be verified; evaluate each new lot in your specific assay context.
