Biological Background: Homeobox protein NANOG Function and Localization
- NANOG (Homeobox protein NANOG) is a transcription regulator essential for inner cell mass and embryonic stem cell proliferation and self-renewal. It imposes pluripotency on ES cells and prevents differentiation towards extraembryonic endoderm and trophectoderm lineages.
- Acts as a transcriptional activator or repressor, binding optimally to DNA consensus sequences 5'-TAAT[GT][GT]-3' or 5'-[CG][GA][CG]C[GC]ATTAN[GC]-3'. Binds to the POU5F1/OCT4 promoter (PubMed:25825768). Related references: PMID:25825768
- Blocks BMP-induced mesoderm differentiation by physically interacting with SMAD1, interfering with coactivator recruitment to SMAD transcriptional complexes.
- When overexpressed, promotes cells to enter S phase and proliferation.
- Contains a homeobox DNA-binding domain and a C-terminal transactivation domain; the protein has a molecular weight of ~34.6 kDa (canonical sequence).

Experimental Guidance and Technical Tips
