Biological Background: NAIP Function and Localization
- NAIP (BIRC1/NLRB1) is a 160 kDa baculoviral IAP repeat-containing protein and member of the NLR family. It functions as an anti-apoptotic factor and a sensor component of the NLRC4 inflammasome.
- It inhibits apoptosis by suppressing the activities of CASP3, CASP7, and CASP9, preventing the autocleavage of pro-CASP9 and cleavage of pro-CASP3, which promotes neuronal survival under pathological conditions.
- Mutations or deletions in NAIP are associated with severe spinal muscular atrophy, as it normally prevents motor neuron apoptosis; it is predominantly expressed in motor neurons and, to a lesser extent, in liver, placenta, and spinal cord.
- In innate immunity, NAIP acts as a specific sensor for bacterial needle protein CprI from Chromobacterium violaceum, triggering NLRC4 inflammasome assembly, caspase-1 activation, cytokine production, and macrophage pyroptosis.
