Biological Background: NAIP Function and Localization
- NAIP (Baculoviral IAP repeat-containing protein 1), also known as neuronal apoptosis inhibitory protein, BIRC1, NLRB1, or psiNAIP, is a member of the inhibitor of apoptosis (IAP) family. It contains three baculoviral IAP repeat (BIR) domains and a nucleotide-binding oligomerization domain (NOD).
- NAIP acts as an anti-apoptotic protein by inhibiting the activities of caspase-3 (CASP3), caspase-7 (CASP7), and caspase-9 (CASP9). It can inhibit the autocleavage of pro-CASP9 and cleavage of pro-CASP3 by CASP9, and it is capable of inhibiting CASP9 autoproteolysis at Asp-315, thus promoting neuronal survival under pathological conditions.
- Mutations or deletions of NAIP are associated with spinal muscular atrophy (SMA), as inappropriate persistence of motor-neuron apoptosis may underlie this disease. NAIP therefore plays a critical role in preventing motor-neuron apoptosis induced by various signals.
- In innate immunity, NAIP functions as a sensor component of the NLRC4 inflammasome. It specifically recognizes and binds needle proteins from pathogenic bacteria such as Chromobacterium violaceum (CprI), leading to inflammasome assembly and activation, caspase-1 activation, cytokine production, and macrophage pyroptosis.
- Expressed predominantly in motor neurons, but not in sensory neurons, NAIP is also found in liver and placenta, and to a lesser extent in the spinal cord.
- NAIP is a multi-functional protein with ATP-binding, metal-binding (zinc), and thiol protease inhibitor activities. It is subject to alternative splicing and is a reference proteome component.
Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide corresponding to residues 1–100 of human NAIP. This may provide a defined epitope useful for competitive ELISA or peptide blocking experiments.
- For immunohistochemistry (IHC-P), consider using tissues with known NAIP expression such as liver, placenta, or motor neuron-rich regions. Antigen retrieval protocols may be necessary and should be optimized for each tissue type.
- In ELISA applications, the antibody may be suitable for both direct and sandwich formats. A positive control using the immunogen peptide can help verify binding specificity.
- Cross-reactivity with mouse NAIP has been reported, making this antibody a potential tool for studies in mouse models of spinal muscular atrophy or bacterial infection. Validate cross-reactivity in the relevant sample system before quantitative conclusions.
- The protein molecular weight is approximately 160 kDa; ensure appropriate gel percentage and transfer conditions for western blot if attempted (note: not validated for WB).
