Biological Background: NADPH oxidase 4 Function and Localization
- NADPH oxidase 4 (NOX4) is a member of the NOX family of NADPH oxidases that primarily catalyzes the reduction of oxygen to hydrogen peroxide (H2O2), and to a lesser extent, superoxide, utilizing NADPH as an electron donor. Related references: PMID:14966267 PMID:15356101 PMID:15927447 PMID:10869423 PMID:11032835 PMID:15155719
- NOX4 functions as an oxygen sensor, regulating the KCNK3/TASK-1 potassium channel and HIF1A activity, and is involved in insulin signaling, apoptosis, bone resorption, and lipopolysaccharide-mediated NFKB activation. Related references: PMID:16019190 PMID:14966267 PMID:15356101 PMID:15572675
- It promotes ferroptosis and reactive oxygen species (ROS) production by activating the NLRP3 inflammasome, and modulates redox signaling in vascular cells through ERK1/2 and ELK1 transcription factor activation, potentially inducing nuclear DNA damage. Related references: PMID:39909992 PMID:15721269 PMID:23393389 PMID:16324151
- Widely expressed with highest levels in kidney, and also present in heart, adipocytes, endothelial cells, skeletal muscle, brain, and airway epithelial cells.
- Localizes to multiple subcellular compartments including the endoplasmic reticulum, cell membrane, focal adhesions, cytoplasm, perinuclear region, nucleus, and nucleolus.
- Contains disulfide bonds, is glycosylated, and possesses transmembrane helices; alternative splicing produces isoforms, some of which lack superoxide-generating activity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 328-578 of human NOX4; consider this region when designing detection strategies or interpreting epitope-specific results.
- For Western blot (WB), a band at approximately 67 kDa is expected; use appropriate protein ladders and validate in kidney lysates as a positive control.
- In immunohistochemistry (IHC-P), optimize antigen retrieval and antibody dilution; note that NOX4 is strongly expressed in kidney distal tubular cells and endothelial cells.
- The antibody shows cross-reactivity with mouse and rat, making it suitable for translational studies; however, verify performance in your specific experimental system.
- For ELISA, pair with a suitable capture antibody, and consider potential interference from recombinant protein constructs.
