Biological Background: NADPH oxidase 4 (NOX4) Function and Localization
- NADPH oxidase 4 (NOX4), also known as KOX-1 or RENOX, is a member of the NADPH oxidase family that primarily catalyzes the reduction of oxygen to hydrogen peroxide (H2O2), and to a lesser extent superoxide (PubMed:14966267, PubMed:15356101, PubMed:15927447, PubMed:10869423, PubMed:11032835, PubMed:15155719). Related references: PMID:14966267, PMID:15356101, PMID:15927447, PMID:10869423, PMID:11032835, PMID:15155719
- It functions as an oxygen sensor, regulating the KCNK3/TASK-1 potassium channel and HIF1A activity (PubMed:16019190). Related references: PMID:16019190
- NOX4 participates in insulin signaling, apoptosis, bone resorption, and lipopolysaccharide-mediated NF-kappa-B activation (PubMed:14966267, PubMed:15356101, PubMed:15572675). Related references: PMID:14966267, PMID:15356101, PMID:15572675
- The enzyme contributes to redox signaling in vascular cells by modulating ERK1/2 and ELK1 transcription factor activation and inducing nuclear DNA damage (PubMed:23393389). Related references: PMID:23393389
- NOX4 promotes ferroptosis and ROS production via NLRP3 inflammasome activation and cytokine release (PubMed:39909992). Related references: PMID:39909992
- It localizes to multiple subcellular compartments including the cytoplasm, endoplasmic reticulum membrane, cell membrane, focal adhesions, nucleus, nucleolus, and perinuclear region.
- Widely expressed, with highest levels in the kidney; also present in heart, adipose tissue, endothelial cells, skeletal muscle, brain, and airway epithelial cells.
- Post-translationally, NOX4 is a glycoprotein with disulfide bonds and transmembrane domains, and undergoes alternative splicing.
Experimental Guidance and Technical Tips
- The recombinant protein immunogen is designed against human NOX4; validate specificity in the relevant species and cell/tissue type.
- For Western blotting, anticipate a band around 67 kDa; be aware of potential post-translational modifications that may shift migration.
- In immunofluorescence/ICC, NOX4 exhibits multiple localizations; appropriate fixation and permeabilization may be required to detect membrane, cytoplasmic, or nuclear pools.
- ELISA performance may depend on the antibody pair and antigen format; consider using recombinant NOX4 protein as a standard.
- Human cross-reactivity is indicated; verify in your experimental system.
