Biological Background: NOX4 Function and Localization
- NADPH oxidase 4 (NOX4) is a multi-pass membrane protein belonging to the NADPH oxidase family. It is also known as kidney oxidase-1 (KOX-1) and renal NAD(P)H-oxidase, with alternative splicing producing multiple isoforms.
- NOX4 primarily catalyzes the reduction of oxygen to hydrogen peroxide (H2O2), and to a lesser extent, superoxide, using NADPH as an electron donor. It acts as an oxygen sensor, regulating the KCNK3/TASK-1 potassium channel and HIF1A activity, and modulates insulin signaling. - NOX4 also contributes to apoptosis, bone resorption, and lipopolysaccharide-mediated NF-kB activation.
