Biological Background: NOX4 Function and Localization

  • NADPH oxidase 4 (NOX4) is a multi-pass membrane protein belonging to the NADPH oxidase family. It is also known as kidney oxidase-1 (KOX-1) and renal NAD(P)H-oxidase, with alternative splicing producing multiple isoforms.
  • NOX4 primarily catalyzes the reduction of oxygen to hydrogen peroxide (H2O2), and to a lesser extent, superoxide, using NADPH as an electron donor. It acts as an oxygen sensor, regulating the KCNK3/TASK-1 potassium channel and HIF1A activity, and modulates insulin signaling. Related references: PMID:14966267, PMID:15356101, PMID:15927447, PMID:10869423, PMID:11032835, PMID:15155719, PMID:16019190
  • NOX4 also contributes to apoptosis, bone resorption, and lipopolysaccharide-mediated NF-kB activation. In the nucleus, it generates superoxide, influencing gene expression and promoting ferroptosis through NLRP3 inflammasome activation and cytokine release. Related references: PMID:15356101, PMID:15572675, PMID:16324151, PMID:39909992
  • An additional isoform or context-dependent activity involves direct superoxide generation, playing a key role in vascular redox signaling, nuclear ERK1/2 activation, and DNA damage induction. Related references: PMID:15721269, PMID:23393389
  • Some splice variants lack superoxide-generating NADPH oxidase activity, indicating functional diversity within the family.
  • Subcellularly, NOX4 localizes to the endoplasmic reticulum membrane, cytoplasm, cell membrane, focal adhesions, nucleus (including nucleolus), and perinuclear region, reflecting its multifaceted signaling roles.
  • Tissue expression is broad, with strong presence in the kidney (distal tubular cells), heart, endothelial cells, skeletal muscle, brain, adipocytes, and airway epithelial cells, as evidenced by both mRNA and protein data.
  • Post-translational modifications include glycosylation and disulfide bonds, consistent with its transmembrane topology and subcellular trafficking.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a region within amino acids 328–578 of human NOX4; ensure that your sample system expresses the full-length or relevant isoform containing this epitope.
  • For Western blot (WB), a band at approximately 67 kDa is expected; use molecular weight markers and validate band specificity with knockout (KO) control lysates when available.
  • In immunohistochemistry on paraffin-embedded sections (IHC-P), antigen retrieval optimization may be necessary, and kidney tissue sections serve as a positive control due to high NOX4 expression.
  • The antibody cross-reacts with mouse and rat NOX4, making it suitable for rodent model studies; however, confirm reactivity in your specific experimental conditions.
  • For ELISA applications, consider pairing with a capture antibody against a different epitope to avoid steric hindrance if using a sandwich format.
