Biological Background: NADPH oxidase 4 (NOX4) Function and Localization
- NADPH oxidase 4 (NOX4) is a multi-pass membrane protein (UniProt keywords: Transmembrane, Transmembrane helix) belonging to the NADPH oxidase family. It is also known as Kidney oxidase-1 (KOX-1), Renal NAD(P)H-oxidase (RENOX), and Superoxide-generating NADPH oxidase 4.

- Functions as an oxygen sensor regulating potassium channels (e.g., KCNK3/TASK-1) and HIF1A activity, and modulates insulin signaling, apoptosis, bone resorption, and NF-kB activation (By similarity).
- Promotes ferroptosis and reactive oxygen species (ROS) production by activating the NLRP3 inflammasome, leading to cytokine release and reduced glutathione levels (By similarity).
