• NACA (Nascent polypeptide-associated complex subunit alpha, also known as alpha-NAC) is a chaperone that binds nascent polypeptide chains as they emerge from the ribosome, preventing inappropriate targeting of non-secretory proteins to the endoplasmic reticulum (ER).
• It blocks the interaction of nascent chains with the signal recognition particle (SRP) and reduces the affinity of ribosomes for ER membrane translocation sites.
• Additionally, NACA acts as a transcriptional coactivator for JUN, binding DNA and stabilizing JUN homodimers at target gene promoters.
• The protein is ubiquitously expressed and localizes to both the cytoplasm and nucleus.
• Post-translational modifications include acetylation, phosphorylation, and ubiquitination, reflecting its regulation in protein transport and transcription.
