Biological Background: NAA50 Function and Localization
- NAA50 (N-alpha-acetyltransferase 50), also known as NAT13, NAT5, SAN, MAK3, and hNaa50p, is an acetyltransferase family member that undergoes alternative splicing.
- It catalyzes N-terminal acetylation of proteins retaining their initiator methionine, showing broad substrate specificity but unable to acetylate peptides with proline in the second position. Related references: PMID:19744929, PMID:21900231, PMID:22311970, PMID:27484799
- NAA50 also displays N-epsilon-acetyltransferase activity, acetylating specific lysine residues (such as histone H4 in vitro) and undergoing autoacetylation in vivo. Related references: PMID:19744929
- It is a component of N-alpha-acetyltransferase complexes with NAA10 and NAA15, where it negatively regulates the NatA complex N-alpha-acetyltransferase activity. Related references: PMID:16507339, PMID:27484799, PMID:29754825, PMID:32042062
- NAA50 is essential for sister chromatid cohesion during mitosis by facilitating CDCA5/sororin binding to cohesin, likely counteracting NAA10 function. Related references: PMID:17502424, PMID:27422821
- The protein localizes to both cytoplasm and nucleus, consistent with its roles in translation-coupled acetylation and chromatin dynamics.
- Post-translational modifications identified by proteomics include acetylation and phosphorylation, contributing to its regulation.
Experimental Guidance and Technical Tips
- For Western blot (WB), expect a band at approximately 19 kDa; use positive controls from human, mouse, or rat cell lines (e.g., HeLa, HEK293, NIH/3T3).
- In immunofluorescence/immunocytochemistry (IF/ICC), optimize fixation and permeabilization; NAA50 signal may be observed in both cytoplasmic and nuclear compartments.
- For ELISA applications, the recombinant immunogen (aa 1–169) can serve as a positive control; note that polyclonal antibodies may exhibit lot-to-lot variability.
- Cross-reactivity with human, mouse, and rat has been reported; verify sequence homology before using on other species.
- Consult public expression databases (e.g., Human Protein Atlas) to select tissues or cell lines with adequate NAA50 expression for your experiments.
