Biological Background: N6AMT1/HEMK2 Function and Localization
- N6AMT1, also known as HEMK2, KMT9, or PrmC, is a methyltransferase that catalyzes methylation of both proteins and, to a lesser extent, arsenic. Related references: PMID:18539146, PMID:21193388, PMID:30017583
- It is the catalytic subunit of a heterodimer with TRMT112, which monomethylates 'Lys-12' of histone H4 (H4K12me1), a mark found at promoters of cell cycle regulator genes. Related references: PMID:31061526
- The enzyme also catalyzes N5-methylation of glutamine residues on proteins containing a Gly-Gln-Xaa-Xaa-Xaa-Arg motif, including the translation termination factor ETF1 on 'Gln-185'. Related references: PMID:18539146, PMID:31632689, PMID:31636962
- In arsenic metabolism, N6AMT1 may mediate the conversion of toxic monomethylarsonous acid into less toxic dimethylarsonic acid, though its role is limited compared to AS3MT. Related references: PMID:21193388, PMID:25997655
- Subcellular localization is primarily nuclear, consistent with its role in histone modification and chromatin regulation.
- It is widely expressed, with highest levels in parathyroid and pituitary glands, followed by adrenal gland and kidney, and lowest in leukocytes and mammary gland.
- Keywords: 3D-structure, Alternative splicing, Chromatin regulator, Methyltransferase, Nucleus, Proteomics identification, Reference proteome, S-adenosyl-L-methionine, Transferase, Ubl conjugation.
Experimental Guidance and Technical Tips
- Immunogen: Recombinant fusion protein covering amino acids 1-214 of human N6AMT1/HEMK2. This full-length immunogen suggests broad epitope recognition, but consider potential cross-reactivity with other methyltransferases sharing structural homology; validate specificity using knockout or knockdown controls.
- For Western blot (WB), endogenous N6AMT1/HEMK2 is predicted at ~23 kDa. Ensure adequate separation by SDS-PAGE and optimize primary antibody dilution for your sample type and detection system.
- For immunohistochemistry (IHC-P), tissues with high expression (e.g., pituitary, adrenal gland) may serve as positive controls, while leukocytes or mammary gland may show weaker staining. Include appropriate antigen retrieval and normal IgG controls.
- ELISA applications may be used for quantitative detection; consider pairing with a detection antibody and optimizing the capture/detection format for your assay system.
