Biological Background: N6AMT1/HEMK2 Function and Localization
- HEMK2 acts as the catalytic subunit in a complex with TRMT112 to monomethylate histone H4 at Lys-12 (H4K12me1), an epigenetic mark found at promoters of numerous cell cycle regulator genes. Related references: PMID:31061526
- It catalyzes N5-methylation of glutamine residues within a Gly-Gln-Xaa-Xaa-Xaa-Arg motif on target proteins, including methylation of translation termination factor ETF1 at Gln-185 when ETF1 is bound to ERF3 in its GTP-bound state. Related references: PMID:18539146 PMID:20606008 PMID:31061526 PMID:31632689 PMID:31636962
- Additionally, HEMK2 may modulate arsenic toxicity by converting monomethylarsonous acid (III) to the less toxic dimethylarsonic acid, although its role in arsenic metabolism is limited compared to AS3MT. Related references: PMID:21193388 PMID:25997655
- The protein is predominantly localized in the nucleus, consistent with its roles in histone modification and gene regulation.
- HEMK2 is widely expressed across human tissues, with highest levels in the parathyroid and pituitary glands, followed by the adrenal gland and kidney; lower expression is observed in leukocytes and mammary gland.
- Keywords associated with HEMK2 include chromatin regulator, methyltransferase, alternative splicing, and S-adenosyl-L-methionine, reflecting its diverse functional and structural characteristics.
Experimental Guidance and Technical Tips
- The immunogen used for this antibody corresponds to the N-terminal sequence (aa 1-186) of human N6AMT1/HEMK2, which may allow detection of full-length protein as well as certain shorter isoforms; validation in the relevant species is recommended.
- For Western Blot (WB), an expected band size of approximately 23 kDa should be monitored; consider optimizing antibody dilution and blocking conditions for specific sample types.
- In immunofluorescence (IF/ICC), a predominant nuclear staining pattern is anticipated based on the known subcellular localization of HEMK2; appropriate nuclear counterstains and negative controls should be included.
- This antibody has been reported to cross-react with mouse and rat, but cross-reactivity in each species and application should be verified experimentally.
